CBX8基因在肝细胞癌中的表达及功能分析

摘要:
[摘要] 目的 探讨色素框同源蛋白8(CBX8)在肝细胞癌(HCC)中的表达情况、临床意义及作用机制,为HCC的预防、诊治和预后提供新思路和方向。方法 基于生物信息学的方法,应用肿瘤基因数据库分析CBX8在HCC组织中的表达和基因突变情况,并分析其与人口学特征、病理特征及预后的关系。利用蛋白质相关数据库分析CBX8基因的蛋白质表达水平,构建相关蛋白质互作网络并进行通路富集分析。结果 HCC组织的CBX8 mRNA表达水平高于正常肝组织。CBX8 mRNA的表达与体重、TP53突变情况、组织学分类、肿瘤分级存在显著关联(P<0.05),且CBX8 mRNA高表达患者的总体生存率较低(P<0.05)。CBX8基因的突变类型主要为C>T突变和错义突变。CBX8主要参与细胞周期、组蛋白泛化等生理过程,可能在干细胞多能性的信号调控通路和癌症关联的microRNA网络中发挥作用。结论 CBX8在HCC中高表达,与HCC预后相关,可能参与了HCC的发生和发展。

Expression and function analysis of CBX8 gene in hepatocellular carcinoma

Abstract:
[Abstract] Objective To explore the expression, clinical significance and mechanism of chromobox 8(CBX8) in hepatocellular carcinoma(HCC), and to provide new ideas and directions for the prevention, diagnosis, treatment and prognosis of HCC. Methods Based on bioinformatics methods, the expression and gene mutation of CBX8 in HCC tissues were analyzed using tumor gene databases, and their relationship with demographic characteristics, pathological characteristics and prognosis of HCC was analyzed. The protein expression level of CBX8 gene was analyzed by protein-related databases, and the related protein interaction network was constructed and the pathway enrichment analysis was carried out. Results The expression level of CBX8 mRNA in the HCC tissues was higher than that in the normal liver tissues. The expression of CBX8 mRNA in the HCC tissues was related to weight, TP53 mutation, histological classification, and tumor stage(P<0.05). The HCC patients with high expression level of CBX8 mRNA had a lower overall survival rate(P<0.05). The mutation types of CBX8 gene were mainly C>T mutation and missense mutation. CBX8 was mainly involved in cell cycle, histone generalization and other physiological processes, and might play a role in signaling pathway to regulate pluripotency of stem cells and cancer-associated microRNA network. Conclusion CBX8 is highly expressed in HCC and is related to the prognosis of HCC, and may be involved in the occurrence and development of HCC.
