miR-27a下调表达对大鼠心肌缺血再灌注后急性肺损伤的保护作用及机制研究

摘要:
[摘要] 目的 探讨miR-27a下调表达对大鼠心肌缺血再灌注(MI/R)后急性肺损伤(ALI)的保护作用及其机制。方法 选择8~12周龄成年雄性SD大鼠40只,将其随机分为假手术组(SP组)、模型组(I/R组)、miR-27a抑制剂组(anta-27a+I/R组)和miR-27a抑制剂空白对照组(NC+I/R组),每组10只。结扎冠脉左前降支30 min,再灌注120 min,构建大鼠MI/R所致ALI模型。anta-27a+I/R组与NC+I/R组在建模前连续3 d分别予尾静脉注射antagomir-27a、antagomir-NC。比较四组大鼠肺脏湿干重比(W/D)及病理改变情况。比较四组肺组织中丙二醛(MDA)、超氧化物歧化酶(SOD)的表达水平。比较四组肺组织及血清中白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的表达水平。结果 与SP组比较,I/R组、anta-27a+I/R组、NC+I/R组大鼠肺组织病理损伤严重,肺W/D值升高;肺组织MDA水平上升,SOD水平降低;肺组织及血清中IL-6、TNF-α水平上升,差异有统计学意义(P<0.05)。与I/R组比较,anta-27a+I/R组大鼠肺组织病理损伤程度有所减轻,肺W/D值下降;肺组织MDA水平下降,SOD水平上升;肺组织及血清中IL-6和TNF-α水平下降,差异有统计学意义(P<0.05)。结论 下调miR-27a表达可以通过减轻炎症反应与氧化应激缓解大鼠MI/R后所引起的ALI。

A study on the protective effects and mechanisms of down-regulated expression of miR-27a on alleviating acute lung injury after myocardial ischemia-reperfusion in rats

Abstract:
[Abstract] Objective To investigate the protective effects and mechanisms of down-regulated expression of miR-27a on alleviating acute lung injury(ALI) after myocardial ischemia-reperfusion(MI/R) in rats. Methods Forty adult male Sprague-Dawley(SD) rats aged 8-12 weeks were randomly divided into sham operation group(SP group), model group(I/R group), miR-27a inhibitor group(anta-27a+I/R group) and miR-27a inhibitor blank control group(NC+I/R group), with 10 rats in each group. The rat model of ALI caused by MI/R was established by ligation of the left anterior descending branch of the coronary artery for 30 minutes and then reperfusion for 120 minutes. Antagomir-27a and antagomir-NC were injected through tail vein in the anta-27a+I/R group and the NC+I/R group for 3 consecutive days before modeling. The wet/dry weight ratio(W/D) and pathological changes of the lung were compared among the four groups. The expression levels of malondialdehyde(MDA) and superoxide dismutase(SOD) in the lung tissues were compared among the four groups. The expression levels of interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in the lung tissues and serum were compared among the four groups. Results Compared with those in the SP group, the lung tissues of rats in the I/R group,the anta-27a+I/R group and the NC+I/R group were seriously injured, and the lung W/D value was increased, and the level of MDA in the lung tissues was increased, and the level of SOD in the lung tissues was decreased, and the levels of IL-6 and TNF-α in the lung tissues and serum were increased, and the differences were statistically significant(P<0.05). Compared with those in the I/R group, the degree of pathological injury of the lung tissues of rats in the anta-27a+I/R group was alleviated, and the lung W/D value was decreased, and the level of MDA in the lung tissues was decreased and the level of SOD in the lung tissues was increased, and the levels of IL-6 and TNF-α in the lung tissues and serum were decreased, and the differences were statistically significant(P<0.05). Conclusion Down-regulation of miR-27a expression can alleviate ALI induced by MI/R in rats by reducing inflammatory response and oxidative stress.
