水通道蛋白3在博莱霉素诱导的硬皮病小鼠真皮组织中的表达

[摘要] 目的 研究博莱霉素(BLM)诱导的硬皮病小鼠真皮组织中水通道蛋白(AQP)3的表达,探讨AQP3在硬皮病中可能的作用机制。方法 将BALB/c小鼠分为PBS组和BLM组,每组各30只,以PBS或BLM皮下注射小鼠背部,1次/d,连续28 d,分别于第7、14、28天注射完毕后处死PBS组及BLM组小鼠各10只,比较皮肤组织中的羟脯氨酸、H2O2和真皮AQP3 mRNA及AQP3蛋白水平。结果 注射28 d后,BLM组和PBS组小鼠皮肤中的羟脯氨酸含量分别为(3028.900±485.021)和(1484.500±247.071)(μg/g湿重),H2O2分别为(30.919±8.278)和(6.357±2.337)(μmol/gprot),真皮AQP3 mRNA分别为(2.090±0.382)和(1.005±0.122),western blots检测真皮AQP3蛋白的相对灰度值为(2.285±0.552)和1,羟脯氨酸、H2O2、AQP3 mRNA及AQP3蛋白在两组之间的差异均有统计学意义(P均<0.05)。结论 AQP3在硬皮病小鼠真皮中表达增高,可能与其转运H2O2的功能有关。

The expression of Aquaporin 3 in the dermal skin of a bleomycin-induced mouse model of scleroderma

Abstract:
[Abstract] Objective To determine the expression of Aquaporin(AQP) 3 in the dermal skin of a bleomycin(BLM)-induced mouse model of scleroderma, and to explore the possible mechanisms of AQP3 in scleroderma.Methods Sixty BALB/c mice were divided into PBS group and BLM group, with 30 cases in each group. PBS or BLM was injected subcutaneously in the backs of the mice once per day for 28 days. Ten mice in each group were sacrificed 7, 14, and 28 days after the injection. Hydroxyproline and hydrogen peroxide(H2O2) in the skin and AQP3 mRNA and AQP3 protein in the dermal skin were compared between the two groups.Results 28 days after the injection, the levels of hydroxyproline in the skin of the BLM group and the PBS group were (3028.900±485.021) and (1484.500±247.071)(μg/g tissue), respectively, and the H2O2 levels were (30.919±8.278) and (6.357±2.337)(μmol/g prot), respectively, and the AQP3 mRNA levels in the dermal skin were (2.090±0.382) and (1.005±0.122), respectively, and the relative gray values of AQP3 protein in the dermal skin detected by Western blots were (2.285±0.552) and 1, respectively. There were significant differences in the levels of hydroxyproline, H2O2, AQP3 mRNA and AQP3 protein between the two groups(P<0.05).Conclusion Higher expression of AQP3 in BLM-treated mice than in PBS-treated mice may be related to its function of transporting H2O2.
