直肠癌放疗敏感性相关miR-93-5p靶基因的预测及其临床意义

摘要:
[摘要] 目的 探索miR-93-5p调控直肠癌放疗敏感性的分子基础,挖掘靶基因并探讨其参与直肠癌发生发展的可能机制及临床意义。方法 从GEO数据库中收集与放疗疗效相关的直肠癌临床数据集,利用limma.R包计算出与放疗敏感性相关的差异表达基因。在多个microRNA靶基因预测数据库中预测miR-93-5p的靶基因。通过对差异表达基因集与靶基因取交集得到PLS1基因。以PLS1的表达水平进行患者的生存分析和基因集富集分析(Gene set enrichment analysis),挖掘PLS1的潜在临床意义及其分子机制。结果 PLS1基因高表达患者的预后明显差于低表达患者(P=0.01)。结论 在直肠癌中PLS1基因高表达是一种预后不良因子,可以作为预测患者预后的有效生物标志物。

Prediction and clinical significance of radiosensitivity gene of miR-93-5p in rectal cancer

Abstract:
[Abstract] Objective To study the prediction and clinical significance of radiosensitivity gene of miR-93-5p in rectal cancer.Methods The data of rectal cancer treated by radiotherapy were collected from the GEO database. Limma.R package software was used to calculate the differentially expressed genes to obtain the first gene set. The target gene of miR-93-5p in multiple micro-RNA target gene prediction database was used to predict and obtain the additional gene set. Plastin 1(PLS1) gene is achieved by intersecting two gene sets. Then survival analysis and gene set enrichment analysis of the expression level of PLS1 were performed to explore the clinical significance of molecular mechanisms of PLS1.Results The total survival time of the patients with higher-level expression of PLS1 was obviously shorter than that of the patients with low-expression PLS1 gene(P=0.01).Conclusion Higher-level expression of PLS1 gene is a bad prognostic factor for rectal cancer and can be used as an effective biomarker to predict the prognosis of the patients.
