MicroRNA-181b相对表达量与冠状动脉狭窄程度的相关性研究

摘要:
[摘要] 目的 探讨冠心病患者外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)中的MicroRNA-181b相对表达量与冠状动脉病变程度的关系,研究MicroRNA-181b对冠脉病变的预测意义。方法 选取因急慢性胸痛在心内科住院且已行冠状动脉造影的患者117例,其中非冠心病组42例,冠心病组75例,根据症状、心电图、肌酸激酶同工酶、肌钙蛋白、冠状动脉造影结果将冠心病组分为稳定型心绞痛30例,不稳定型心绞痛22例和急性心肌梗死23例。采集各组患者一般临床资料,抽血检测相关生化指标,计算冠脉Gensini积分,采用荧光实时定量PCR方法检测MicroRNA-181b的相对表达量,研究MicroRNA-181b相对表达量与Gensini积分有无关系,探讨其在冠心病中的意义。结果 冠心病组Gensini积分与MicroRNA-181b表达水平呈负相关。冠心病组MicroRNA-181b相对表达量均低于非冠心病组,差异有统计学意义(P<0.05)。结论 冠心病患者MicroRNA-181b表达水平降低,表达水平越低其冠脉病变程度(Gensini积分)越重,呈负相关,MicroRNA-181b高表达对冠状动脉粥样硬化的进展有抑制作用。

Abstract:
[Abstract] Objective To investigate the correlation between relative expression of MicroRNA-181b and the severity of coronary artery lesions in peripheral blood mononuclear cells(PBMCs) of the patients with coronary artery disease(CHD), and to study the significance of modification of MicroRNA-181b in patients with coronary artery disease. Methods A total of 117 patients with chest pain who received coronary angiography in department of cardiology of our hospital were selected, including 42 cases in the control group and 75 cases in the coronary heart disease(CHD) group. According to the symptoms, ECG, CK-MB, troponin and the findings in coronary angiography, the CHD group was divided into three different subgroups: stable angina subgroup(n=30, SAP subgroup), unstable angina subgroup(n=22, UAP subgroup) and acute myocard infarction subgroup(n=22, AMI subgroup). In each group, the general clinical data, blood biochemical markers, as well as Gensini scores were recorded. The relative expression of MicroRNA-181b was detected by real-time fluorescent quantitative PCR. To explore the possibility of application of microRNA-181b in coronary artery diseas. Results In the CHD group, Gensini scores were negatively correlated with the relative expression level of MicroRNA-181b. The relative expression levels of MicroRNA-181b in the CHD group were significantly lower than those in the control group(P<0.05). Conclusion In coronary heart disease group, the relative expression of microRNA-181b decreased, the lower relative expression of microRNA-181b the heavier severity of coronary artery lesions. The relative expression levels of microRNA-181b was negatively correlated with the severity of coronary artery stenosis.Higher relative expression of microRNA-181b is likely to be a inhibitory factor in the development of coronary heart disease.
