[摘要] 目的 检测miR-155-5p在鼻咽癌组织中的表达水平,探讨miR-155-5p与远处转移及临床预后的关系。方法 收集有远处转移(转移组)和仅有原发病灶(非转移组)鼻咽癌活检标本共60例,应用实时荧光定量-PCR(QRT-PCR)方法检测鼻咽癌中miR-155-5p的表达水平,分析miR-155-5p与临床病理参数及预后的关系。结果 远处转移组miR-155-5p表达水平较非转移组显著上调,相对表达量为[(4.92±5.52) vs (1.98±2.66)](t=-2.505,P=0.017)。miR-155-5p表达水平与性别、年龄、发病部位、淋巴结转移、T分期及临床分期等均无明显的相关关系(P>0.05)。以miR-155-5p相对表达量中位值2-△△Ct=1.66为截断值,Kaplan-Meier生存曲线和Log-rank检验结果显示,miR-155-5p高表达组鼻咽癌患者5年总生存率明显低于低表达组(χ2=4.18,P=0.041)。结论 miR-155-5p表达水平与鼻咽癌远处转移密切相关,可能成为辅助判断患者预后的生物学指标。 |

[Abstract] Objective To investigate the expression of miR-155-5p in nasopharyngeal carcinomas(NPC) and its relationship between metastasis and prognosis in NPC patients. Methods The biopsy specimens of 60 NPC patients with distant metastasis(metastatic group) or primary cancer focus alone(non-metastatic group) in our hospital were collected, and the expressions of miR-155-5p in the specimens were detected by real time fluorescent quantitative PCR(QRT-PCR). The relationship between miR-155-5p and the clinicopathological parameters and prognosis were analyzed. Results Compared with that in the non-metastatic group,the level of miR-155-5p in the metastatic group was significantly up-regulated[(4.92±5.52) vs (1.98±2.66)](t=-2.505,P=0.017). No significant differences were observed among miR-155-5p and sex,age,location, lymph node metastasis, T grades and clinical stages(P>0.05). If the median level of miR-155-5p was considered as cut-off value, Kaplan - Meier survival curve and the Log-rank test results showed that the overall 5 year survival rate of the NPC patients with low expression of miR-155-5p was significantly higher than that of the NPC patients with high expression of miR-155-5p(χ2=4.18, P=0.041 ). Conclusion The expression level of miR-155-5p may play a key role in metastasis of NPC. It may be a novel biomarker in predicting prognosis of NPC. |
