实时荧光定量PCR检测TMPRSS4 mRNA在41例肺腺癌中的表达及临床意义

摘要:
[摘要] 目的 探讨通过实时荧光定量PCR法检测临床切除的肺腺癌、癌旁组织及正常肺组织中的Ⅱ型跨膜丝氨酸蛋白酶(TMPRSS4)mRNA的表达与非小细胞肺腺癌患者关系。方法 采用RQT-PCR法检测41例非小细胞肺腺癌及其配对癌旁组织和12例良性疾病肺组织中TMPRSS4 mRNA表达情况。结果 非小细胞肺腺癌组织TMPRSS4 mRNA表达明显高于配对癌旁组织(P<0.01),TMPRSS4 mRNA在配对癌旁组织的相对表达量显著高于正常肺组织(P<0.01)。TMPRSS4 mRNA表达与非小细胞肺腺癌淋巴结转移、临床分期相关(P<0.05);与患者年龄、性别及病理分化程度无关。结论 TMPRSS4在非小细胞肺腺癌中的表达与肿瘤发生、转移密切相关,高表达的TMPRSS4 mRNA提示肺腺癌有转移风险更高及预后更差的可能,检测TMPRSS4 mRNA有助于提高非小细胞肺腺癌恶性程度的判断。

Abstract:
[Abstract] Objective To explore the expressions of TMPRSS4 mRNA in lung adenocarcinoma tissues and their correlations with lung adenocarcinoma.Methods The expressions of TMPRSS4 mRNA in cancerous tissues of 41 patients with lung adenocarcinoma, and their matched adjacent tissues, and the expressions of lung tissues of 12 cases with benign pulmonary disease were detected using real-time fluorescent quantitative PCR.Results The expressions of TMPRSS4 mRNA in lung adenocarcinoma tissues were higher than those in the matched adjacent tissues(P<0.01). The expressions of TMPRSS4 mRNA in the normal lung tissues were lower than those in the matched adjacent tissues(P<0.01). There was significant correlation between the expression of TMPRSS4 mRNA and the lymph node metastasis, and TNM stage of lung adenocarcinoma(P<0.05). There was no significant correlation between the expression of TMPRSS4 mRNA and age, gender and the degree of pathological differentiation.Conclusion TMPRSS4 expression in lung adenocarcinoma is closely related to the tumor occurrence and metastasis. Higher expression of TMPRSS4 mRNA indicates higher risk of tumor metastasis and the possibility of a worse prognosis in the patients with lung adenocarcinoma.
