P2X7受体在缺血性中风发病中的作用研究进展

摘要:
缺血性中风是脑缺血的最突出和急性表现,是一种以脑血流中断引起神经系统迅速损伤的综合征[1]。脑缺血后,位于缺血中心区的脑血流量显著降低,导致快速能量衰竭,ATP消耗,离子失衡和神经元死亡[2]。细胞外ATP在中枢神经系统中是介导不同类型细胞之间相互作用的重要的信号分子[3],它是一种重要的神经递质,通过激活嘌呤能受体P2X和P2Y发挥多种作用。P2X7R是嘌呤能受体P2X家族中非常独特的一个亚类,可能处于神经信号过程中相当上游的调节位点,通过调节递质释放、炎症反应、细胞凋亡和增殖,在神经损害及随后的修复过程中起重要作用[4]。被认为是缺血性中风、阿尔茨海默病、多发性硬化症、脊髓损伤、神经性疼痛等疾病潜在的治疗靶点[5]。笔者就近年来P2X7R在缺血性中风的作用进行总结。

Abstract:
Ischemic stroke is the most prominent and acute manifestation of cerebral ischemia. It is a syndrome of rapid damage of nervous system caused by interruption of cerebral blood flow. After cerebral ischemia, cerebral blood flow in the ischemic center decreased significantly, resulting in rapid energy failure, ATP consumption, ion imbalance and neuronal death < sub > < sup > [2] < / sup > < / sub >. Extracellular ATP is an important signaling molecule < sub > < sup > [3] < / sup > < / sub > that mediates the interaction between different types of cells in the central nervous system. It is an important neurotransmitter and plays a variety of roles by activating purinergic receptors P2X and P2Y. P2X7R is a very unique subclass of the purinergic receptor P2X family. It may be located at a fairly upstream regulatory site in the neurosignaling process. P2X7R plays an important role in neurological damage and subsequent repair by regulating the release of neurotransmitters, inflammatory response, apoptosis and proliferation. It is considered as a potential therapeutic target for ischemic stroke, Alzheimer's disease, multiple sclerosis, spinal cord injury, neuropathic pain and other diseases < sub > < sup > [5] < / sup > < / sub >. The author summarized the role of P2X7R in ischemic stroke in recent years.
