隐丹参酮等中药单体对SZ95细胞内脂质合成的影响

摘要:
[摘要] 目的 观察隐丹参酮等中药单体对SZ95细胞中脂质合成的影响。方法 用尼罗河红荧光染色结合流式细胞术,以异维A酸为对照,观察隐丹参酮、丹参酮II-A、盐酸小檗碱、黄芩甙、苦参碱作用SZ95 细胞48 h后,1×104个尼罗河红染色的SZ95细胞中平均每个细胞的荧光强度变化。结果 1.25×10-7M丹参酮II-A使尼罗河红染色荧光强度下降最为明显(-34.3%,P<0.05),其次为1×10-4M黄芩甙(-30.1%,P<0.05)、1×10-7M异维A酸(-21.0%,P<0.05)、1.25×10-7M隐丹参酮(-16.2%)、1×10-6M盐酸小檗碱(-7.8%)也有抑制作用,但没有统计学差异。1×10-3M苦参碱则显示了促进脂质合成的作用(26.9%)。结论 丹参酮II-A、黄芩甙及异维A酸可能具有直接抑制皮脂腺细胞中脂质合成的作用。

Abstract:
[Abstract] Objective To investigate the effects of Chinese medicines cryptotanshinone, tanshinone II-A, berberine, baicalin, matrine on the lipid synthesis of SZ95 sebocytes compared with isotretinoin.Methods Lipid contents in the SZ95 sebocytes were labeled with a fluorescence dye(Nile red) and analyzed using flow cytometry.Results Lipid synthesis was inhibited by 1.25×10-7M tanshinone II-A(-34.2%), 1×10-4M baicalin(-30.1%), 1×10-7M isotretinoin(-21.1%), 1.25×10-7M Cryptotanshinone(-16.2%)and 1×10-5 M Berberine(-7.8%)also decreased lipogenesis but no statistical significance was found. Matrine increased lipogenesis(26.913%).Conclusion Tanshinone II-A and baicalin may have a direct effect on lipid synthesis in sebocytes.
