| [摘要] 目的 探讨成功建立帕金森病(Parkinson′s disease,PD)大鼠模型的快速有效方法。方法 将Wistar大鼠92只,随机分为实验组(80只)及对照组(12只),采用脑立体定向术,实验组在大鼠右侧中脑腹侧被盖区(ventral tegmental area,VTA)注入6-OHDA,经阿朴吗啡(Apomorphine,APO)诱导表现为恒定左侧旋转且旋转圈数﹥210 r/30 min的视为成功PD大鼠模型,对照组则在脑部相应位置注入生理盐水。免疫组化法观察成功模型毁损侧黑质酪氨酸羟化酶(TH)阳性神经元的形态及数量变化。结果 (1)行为学检测:模型组与对照组手术后分别死亡2只及1只,实验组78只大鼠中36只左侧恒定旋转圈数﹥210 r/30 min,造模成功率为46.2%,并且维持时间长。(2)免疫组化:成功大鼠模型毁损侧黑质区TH阳性的神经元较对侧及对照组明显减少(P<0.01)。结论 大鼠中脑VTA单点注射6-OHDA制作PD大鼠模型,易于定位,操作简单,动物死亡率低,模型成功率较高,成本较低,是较快建立稳定PD大鼠模型的可行方法。 |

| [Abstract] Objective To explore a quick and effective method for the establishment of rat model of Parkinson disease.Methods A total of 92 Wistar rats were randomly divided into experimental group(n=80) and control group(n=12),6-OHDA was injected into rats′ dextral midbrain above the ventral tegmental area(VTA) by stereotaetie technology and apomorphine(APO) was given by intraperitoneal injection on 1,2,3 and 4 weeks after 6-OHDA injection.The rat which showed consistently left whirling and whose numbers of rotation were more than 210 r/30 min induced by APO was judged as PD model.The contral group was injected with saline instead of 6-OHDA.The morphological changes and the number of tyrosine hydroxylase(TH) positive neurons were detected by immunohistoehemistry.Results (1)One rat in the control group and 2 rats in the model group died respectively after the operation.Behavioral test showed that there were 36 from 78 rats regarded as successful PD models whose numbers of rotation were more than 210 r/30 minutes after operation and the rotation was stable,the successful ratio was 46.2%.(2)TH positive neurons of injury VTA in model group were less than those in the opposite side and in control group obviously(P<0.01).Conclusion The one-spot injection method for PD is more convenient and practical with lower mortality of animal and higher successful rate of model establishment,the cost is more low.The stable PD rat models can soon be established. |
