不同浓度二甲双胍对子宫内膜癌细胞增殖与凋亡的影响

摘要:
[摘要] 目的 探讨二甲双胍对子宫内膜癌细胞系Ishikawa细胞增殖、凋亡和细胞周期的影响。方法 以不同浓度及时间二甲双胍作用Ishikawa细胞,采用四甲基偶氮唑蓝比色法和流式细胞仪检测细胞增殖、凋亡和细胞周期。结果 二甲双胍抑制子宫内膜癌细胞增殖,并与作用时间浓度相关。流式细胞仪检测提示二甲双胍作用下细胞中G0/G1期细胞比例增加,S期细胞比例降低。结论 二甲双胍能抑制子宫内膜癌Ishikawa细胞增殖,使细胞生长周期停滞。

Effect of metformin on proliferation and apoptosis of human endometrial carcinona cells

Abstract:
[Abstract] Objective To investigate the effect of metformin on proliferation, cell cycle distribution and apoptosis of human endometrial carcinoma cell lines in vitro.Methods The effects of metformin on proliferation, cell cycle distribution and apoptosis of Ishikawa cell lines were measured by MTT assay and fluorescence-activated cell sorting technique.Results Metformin potently inhibited growth in a dose-dependent manner in Ishikawa cell lines in a dose- and time- dependent fashion. Flow cytometric analysis showed that metformin increased the number of cells in G0/G1 and reduced the percentage of cells in S phase.Conclusion Metformin is a potent inhibitor of cell proliferation in endometrial cancer cell lines.
