肛周脓肿组织中Foxp3、IL-15表达情况及其与疾病复发的关联性研究

摘要:
[摘要] 目的 探讨肛周脓肿组织中叉头样转录因子3(Foxp3)和白细胞介素-15(IL-15)的表达情况,分析其与疾病复发的关联性。方法 选取2019年1月至2020年7月于北京中医医院顺义医院进行手术治疗的肛周脓肿患者98例,收集患者术中的脓肿壁组织以及脓肿壁周围的正常组织。采用免疫组织化学染色法检测组织中Foxp3和IL-15的表达情况。分析肛周脓肿组织Foxp3及IL-15表达与患者临床病理特征和疾病复发的关联性。结果 Foxp3蛋白主要表达于细胞核区域,IL-15蛋白主要表达于胞质和胞膜。脓肿壁组织中Foxp3、IL-15蛋白的阳性表达率高于正常组织,差异有统计学意义(P<0.05)。脓肿壁组织中Foxp3、IL-15表达情况与与脓肿直径、愈合时间和术前肛瘘形成情况具有显著关联性(P<0.05),与性别、年龄、脓肿位置和病程无显著关联(P>0.05)。Cox回归分析结果显示,术前肛瘘、Foxp3阳性、IL-15阳性是促进肛周脓肿患者疾病复发的独立危险因素(P<0.05)。结论 肛周脓肿组织中Foxp3和IL-15表达升高,与患者临床病理特征及疾病复发密切相关。

A study on the expressions of Foxp3 and IL-15 in perianal abscess tissues and their correlations with disease recurrence

Abstract:
[Abstract] Objective To study the expressions of foxhead box protein p3(Foxp3) and interleukin-15(IL-15) in perianal abscess tissues and their correlations with disease recurrence. Methods Ninety-eight patients with perianal abscess who underwent surgical treatment in Shunyi Hospital of Beijing Hospital of Traditional Chinese Medicine from January 2019 to July 2020 were selected. The intraoperative abscess wall tissues and normal tissues around the abscess wall of the patients were collected. The expressions of Foxp3 and IL-15 were detected by immunohistochemical staining. The correlations of Foxp3 and IL-15 expressions in the perianal abscess tissues with the patients′ clinicopathological features and disease recurrence were analyzed. Results Foxp3 was mainly expressed in the cell nucleus region, and IL-15 protein was mainly expressed in the cytoplasm and cell membrane. The positive expression rates of Foxp3 and IL-15 protein in the abscess wall tissues were higher than those in the normal tissues, and the differences were statistically significant(P<0.05). The expressions of Foxp3 and IL-15 in the abscess wall tissues were significantly correlated with abscess diameter, healing time and preoperative anal fistula formation(P<0.05), but were not significantly correlated with gender, age, abscess location and course of disease(P>0.05). The results of Cox regression analysis showed that preoperative anal fistula, Foxp3 positive and IL-15 positive were independent risk factors for disease recurrence in patients with perianal abscess(P<0.05). Conclusion The expressions of Foxp3 and IL-15 are increased in perianal abscess tissues, which is closely related to the patients′ clinicopathological features and disease recurrence.
