川芎嗪对酒精性股骨头缺血性坏死骨细胞凋亡影响的实验研究

摘要:
[摘要] 目的 观察不同剂量川芎嗪对骨细胞凋亡率的影响和对骨组织的保护作用。方法 将实验组48只雄性家兔采用灌胃法,给予烈性普通白酒(含体积分数50%乙醇)8 ml/(kg·d),将实验组随机分成生理盐水对照组和3种不同剂量[8 mg/(kg·d),4 mg/(kg·d),2 mg/(kg·d)]的盐酸川芎嗪干预组。另12只雄性家兔设为正常对照组不做任何处理。以上五组均用药观察至处死为止。于1、2、3、6个月分批处死,用TUNEL法检测各组间骨细胞凋亡情况。结果 第3、6个月时大、中剂量川芎嗪组与生理盐水对照组相比较骨细胞凋亡数量逐渐减少,差异有统计学意义(P<0.05);第6个月时大、中剂量川芎嗪组与正常组相比较骨细胞凋亡数量已较为接近(P>0.05)。结论 一定剂量范围、一定疗程的川芎嗪具有抑制骨细胞凋亡的作用。

Experiment on the influence of ligustrazine on osteocyte apoptosis in alcohol-related necrosis of the femoral head

Abstract:
[Abstract] Objective To observe the effect of ligustrazine on osteocyte apoptosis rate in rabbits,and to explore its protective effect.Methods Forty-eight New Zealand white rabbits were chosen and randomly divided into four groups. The experimental group was administered the strong distilled spirits containing 50%(v/v) of alcohol at a dosage of 8 ml/(kg·d). The experiment groups were divided into Saline control group, and high-,middle- and low-dosage ligustrazine [8,4 and 2 mg/(kg·d) respectively] groups. Moreover 12 New Zealand white rabbits as a normal control group received no treatment. The animals in five groups were sacrificed in batches 1, 2, 3 and 6 months after the procedures. The changes of histology in the Hepar and femoral head were observed. The osteocytes apoptosis was measured by means of TUNEL.Results At 3, 6 months, compared with the control group the number of osteocyte apoptosis in the high-,middle-dosage ligustrazine group gradually reduced. The apoptosis rate of osteocyte differed from those in other groups(P<0.05). At six months,the number of osteocyte apoptosis in high-, middle-dosage ligustrazine groups were close to the number of osteocyte apoptosis in the control group(P>0.05).Conclusion A certain range of dosage and course of ligustrazine has a protective effect on the apoptosis of osteocytes.
