甲强龙对实验性变态反应性脑脊髓炎小鼠的肿瘤坏死因子α和白介素17的影响

摘要:
[摘要] 目的 探讨甲强龙对实验性变态反应性脑脊髓炎(EAE)小鼠的肿瘤坏死因子α(TNF-α)和白介素17(IL-17)的影响。方法 将30只雌性C57BL/6J小鼠随机分为甲强龙组、EAE组和正常组,每组10只。EAE模型通过以髓鞘少突胶质细胞糖蛋白多肽35-55(MOG35-55)为抗原诱导。采用蛋白质印迹法(Western Blot)检测小鼠大脑的TNF-α和IL-17表达水平。结果 甲强龙组的临床症状较EAE组改善(P<0.05),大脑的TNF-α和IL-17表达水平明显低于EAE组(P<0.05)。结论 甲强龙能改善EAE小鼠的症状,其作用机制之一是抑制促炎性因子TNF-α和IL-17的表达。

Effect of methylprednisolone on the levels of TNF-α and IL-17 in mice with experimental autoimmune encephalomyelitis

Abstract:
[Abstract] Objective To explore the effects of methylprednisolone on the levels of TNF-α and IL-17 in the central nervous system of mice with experimental autoimmune encephalomyelitis(EAE).Methods 30 female C57BL/6J mice were randomly divided into 3 groups: methylprednisolone-treated group, EAE group and normal group. The EAE model was induced by immunizing mice with myelin oligodendrocyte glycoprotein peptides(MOG35-55). The brain tissues were collected and their expressions of TNF-α and IL-17 were detected using Western Blot.Results The neurological deficits of the methylprednisolone-treated mice were lighter than those of the EAE mice(P<0.05), while the methylprednisolone-treated mice displayed the lower expressions of TNF-α and IL-17 than the EAE mice did(P<0.05).Conclusion Methylprednisolone can protect against EAE in mice by the inhibition of TNF-α and IL-17.
