白藜芦醇通过抑制NF-κB活性减轻大鼠肠缺血再灌注继发性肺损伤的实验研究

摘要:
[摘要] 目的 评价白藜芦醇(resveratrol,Res)预处理对大鼠肠缺血再灌注继发肺损伤时NF-κB活性的影响。方法 选健康清洁级雌性SD大鼠24只,体重200~220 g,采用随机数字表法分为3组(n=8):假手术组(S组)、肠缺血再灌注组(M组)和白藜芦醇预处理组(R组)。采用夹闭肠系膜上动脉75 min后再灌注4 h的方法制备肠缺血再灌注继发性肺损伤(IIR-ALI)模型。于模型制备前5 d,R组每天腹腔注射Res 15 mg/kg(采用生理盐水稀释),S组和M组腹腔注射等容量生理盐水。于再灌注4 h时放血处死大鼠,收集肺组织和支气管肺泡灌洗液标本,测肺湿干重比值(W/D)、支气管肺泡灌洗液蛋白浓度、支气管肺泡灌洗液中IL-1β和TNF-α含量以及光镜下行肺损伤评分(Hofbauer评分);采用免疫印迹检测肺组织核蛋白NF-κB p65的表达。结果 与S组比较,M组和R组大鼠肺组织损伤严重,肺组织损伤评分、肺W/D值、支气管肺泡灌洗液蛋白浓度、支气管肺泡灌洗液蛋白浓度IL-1β和TNF-α含量以及肺组织核蛋白NF-κB p65的表达增加(P<0.05);与M组比较,R组大鼠肺组织损伤减轻,肺组织损伤评分、肺W/D值、支气管肺泡灌洗液蛋白浓度、支气管肺泡灌洗液蛋白浓度IL-1β和TNF-α含量以及肺组织核蛋白NF-κB p65表达下降(P<0.05)。结论 白藜芦醇预处理可减轻大鼠肠缺血再灌注继发性肺损伤,机制可能与其抑制肺组织NF-κB活性有关。

Resveratrol preconditioning attenuates acute lung injury induced by intestinal ischemia-reperfusion in rats by inhibiting NF-kappaB activity

Abstract:
[Abstract] Objective To evaluate the effect of resveratrol preconditioning on nuclear factor of kappa B(NF-kappaB) activity during acute lung injury induced by intestinal ischemia-reperfusion(IIR) in rats.Methods Twenty-four healthy female SD rats were randomly divided into three groups: sham-operated group(S group, n=8), intestinal ischemia-reperfusion mode group(M group, n=8), resveratrol pretreatment group(R group, n=8). Acute lung injury induced by intestinal ischemia-reperfusion(IIR-ALI) was conducted by clamping the superior mesenteric artery(SMA) in rats for 75 minutes followed by 4 hours′ reperfusion. In the R group, rats were treated with resveratrol(15 mg/kg, intraperitoneally) for 5 days before IIR. In the other groups, rats were treated with the same volume of NS for 5 days before operation. All rats were sacrificed after 4 hours′ reperfusion of SMA. The lung tissues were obtained for calculation of wet/dry weight ratio(W/D) and pathological investigation(Hofbauer′s scoring system). The expression of nucleoprotein NF-kappaB p65 was detected using Western blotting and the bronchoalveolar lavage fluid was collected to detect the total protein content and the levels of TNF-α and IL-1β.Results Compared with those in the S group, the Hofbauer scores, W/D, NF-kappaB p65 expression and BALF indexes including the total protein content and the levels of TNF-α and IL-1β in the M group and the R group were increased significantly(P<0.05). Compared with those in the M group, the Hofbauer scores, W/D, NF-kappaB p65 expression of lung tissue and BALF indexes in the R group were decreased significantly(P<0.05).Conclusion Resveratrol preconditioning can attenuate the acute lung injury induced by intestinal ischemia-reperfusion in rats. This effect may be related to inhibiting NF-kappaB activity in the lungs.
