铜死亡在骨肉瘤中的研究进展

[摘要] 骨肉瘤(OS)起源于间充质干细胞,是最常见的原发性恶性骨肿瘤,通常发生在股骨远端干骺端、胫骨近端和肱骨近端。OS恶性程度高,预后极差,严重威胁儿童和青少年的健康和生命。OS的病理特征是肿瘤细胞形成未成熟的骨样基质,然而其关键病理机制尚未完全阐明。近年来研究表明,通过药理学降低肿瘤内铜水平,可增强抗肿瘤免疫治疗效果。铜死亡是一种铜依赖性的新型细胞程序性死亡方式,由过度堆积的过氧化脂质诱导发生。铜死亡涉及铜的依赖、脂酰化蛋白的积累以及铁硫簇蛋白的减少等过程,在OS的发生与转归中起到了重要的调节作用。该文对铜死亡在OS中的研究进展作一综述。

Research progress of cuprotosis in osteosarcoma

Abstract:
[Abstract] Osteosarcoma(OS) originates from mesenchymal stem cells and is the most common primary malignant bone tumor, typically occurring in the distal femoral metaphysis, proximal tibia and proximal humerus. OS has a high degree of malignancy and a very poor prognosis, seriously threatening the health and life of children and adolescents. The pathological feature of OS is that tumor cells form an immature bone-like matrix, yet its key pathological mechanism has not been fully elucidated. In recent years, studies have shown that reducing copper levels in tumors through pharmacology may enhance the immunotherapy efficacy of the fight against cancer. Cuprotosis is a novel type of copper-dependent programmed cell death, induced by excessive accumulation of peroxidized lipids. Cuprotosis involves processes such as copper dependence, accumulation of fatty-acylated proteins, and reduction of iron-sulfur cluster proteins. These processes play important regulatory roles in the occurrence and outcome of OS. In this paper, the research progress of cuprotosis in OS is reviewed.
