| [摘要] 目的 探讨癫痫患者血清miR-128-3p和miR-211-5p水平,分析其与脑电图(EEG)异常程度的相关性。方法 招募2022年3月至2023年6月青岛市城阳区人民医院收治的癫痫患者120例作为癫痫组,另选择同期健康体检者120名作为对照组。根据EEG检查结果将癫痫患者进一步分为轻度异常组(41例)、中度异常组(46例)和重度异常组(33例)。通过实时荧光定量聚合酶链反应(qRT-PCR)法检测血清miR-128-3p和miR-211-5p水平。通过Spearman秩相关分析血清miR-128-3p和miR-211-5p水平与EEG异常程度的相关性。采用受试者工作特征(ROC)曲线分析血清miR-128-3p和miR-211-5p对EEG重度异常的诊断效能。结果 癫痫组血清miR-128-3p、miR-211-5p水平显著低于对照组(P<0.05)。在癫痫患者中,重度异常组和中度异常组血清miR-128-3p、miR-211-5p水平显著低于轻度异常组(P<0.05),且重度异常组水平较中度异常组更低,差异有统计学意义(P<0.05)。Spearman秩相关分析结果显示,血清miR-128-3p、miR-211-5p水平与EEG异常程度均呈负相关(rs=-0.484,P<0.001;rs=-0.579,P<0.001)。ROC曲线分析结果显示,血清miR-128-3p、miR-211-5p水平均可有效对EEG重度异常进行诊断(P<0.05),且两指标联合的诊断效能较单一指标更高(Z两指标联合-miR-128-3p=2.850,P=0.004;Z两指标联合-miR-211-5p=2.562,P=0.010),其灵敏度为81.82%,特异度为79.31%。结论 癫痫患者血清miR-128-3p和miR-211-5p水平降低,与EEG异常程度呈负相关,两指标联合可以更好地诊断患者EEG重度异常。 |

| [Abstract] Objective To explore the levels of serum microRNA-128-3p(miR-128-3p) and microRNA-211-5p(miR-211-5p) in epilepsy patients and to analyze their correlations with the the degree of electroencephalogram(EEG) abnormalities. Methods A total of 120 epilepsy patients admitted to Qingdao Chengyang People′s Hospital from March 2022 to June 2023 were recruited as epilepsy group, and other 120 healthy individuals who underwent physical examination during the same period were selected as control group. According to the EEG examination results of these epilepsy patients, they were further divided into mild abnormality group(41 patients), moderate abnormality group(46 patients), and severe abnormality group(33 patients). The levels of serum miR-128-3p and miR-211-5p were determined by using quantitative real-time fluorescence polymerase chain reaction(qRT-PCR). The correlations of serum miR-128-3p and miR-211-5p levels with the degree of EEG abnormalities were analyzed by using Spearman rank correlation. The receiver operating characteristic(ROC) curve was used to analyze the efficacy of serum miR-128-3p and miR-211-5p levels in diagnosing severe EEG abnormality. Results The levels of serum miR-128-3p and miR-211-5p in the epilepsy group were significantly lower than those in the control group(P<0.05). The levels of serum miR-128-3p and miR-211-5p in the epilepsy patients of the severe abnormality group and the moderate abnormality group were significantly lower than those in the epilepsy patients of the mild abnormality group(P<0.05), and the levels of miR-128-3p and miR-211-5p in the severe abnormality group were lower than those in the moderate abnormality group, with statistically significant differences(P<0.05). The results of Spearman rank correlation analysis showed that the levels of serum miR-128-3p and miR-211-5p were negatively correlated with the degree of EEG abnormalities(rs=-0.484, P<0.001; rs=-0.579, P<0.001). The results of ROC curve analysis showed that the levels of serum miR-128-3p and miR-211-5p could effectively diagnose severe EEG abnormality(P<0.05), and the diagnostic efficacy of the combination of the two indicators was higher than that of a single indicator(Zcombination of the two indicators-miR-128-3p=2.850, P=0.004; Zcombination of the two indicators-miR-211-5p=2.562, P=0.010), with a sensitivity of 81.82% and a specificity of 79.31%. Conclusion The levels of miR-128-3p and miR-211-5p in the serum of epilepsy patients are decreased, which are negatively correlated with the degree of EEG abnormalities. The combination of the two indicators can better diagnose severe EEG abnormality in the patients. |
